Not a mine?
There is a Pegomya species that mines vining Fallopia spp., but from what I can see I'm not convinced this is a mine.

 
The only thing that gives me
The only thing that gives me pause is on the underside of the leaf, where it looks like a slightly raised lighter green area along the 'trail.' How would I check this out next time? Would cutting across the leaf with an X-acto blade show if it's a mine?

 
That should work, yes
I see what you're talking about and I don't have a good explanation for it. The species normally found on this host would have an eggshell (or more typically several parallel eggshells) on the underside of the leaf at the beginning of the mine--or else an obvious entry hole made by an older larva.
